- Work Clean – Use a flow hood or still air box.
- Inoculate – Transfer culture, swab, clone, or spores using sterile tools.
- Incubate – Store at 68–78°F until colonies appear.
- Seal & Label – Wrap with Parafilm or grafting tape.
